The allopregnanalone theory. This could be it.

I’ve considered there to be a problem with neurosteriods from the beginning, particulary allopregnanalone, this undoubtedly would be the cause of brain fog and emotional numbness. However I’m not convinced this is our only problem… I’ve seen a few articles indicating low levels of allopregnanalone could be the cause of MS and Alzheimer’s…

Stem cell treatment could be a good option for us. If I was able to get my emotions back I’d be happy enough.

For sure neurosteroids are responsible for the mental side effects. But I wonder about the sexual side effects too. How is it that post SSRI sufferers have such similar symptoms? And its most likely the SSRI drug screwed up something in the brain. MDMA has been shown to cause similar symptoms as well.

Obviously this study is not of the best participant field, but interesting that this drug, metformin, which is an antidiabetic drug restored “normal allopregnanolone levels”. The kicker is it reduces Testosterone and LH (perhaps it increase 5ar2 activity, which would increase the conversion rate of T to DHT). Perhaps testosterone supplementation will also be required.

For me it would appear this drug targets the right place for us. We know we have something wrong with our allopregnanolone levels, we know there is no 5-alpha reductase type 2 in the brain (so in theory finasteride didn’t affect brain production), therefore the low level of allopregnanolone must be coming away from the brain, such as gonads, adrenals.

ncbi.nlm.nih.gov/pubmed/20624011

Nice find Tim, what’s your take on reduced allopregnanalone levels possibly increasing its precursor progesterone since its synthesis is inhibited. The adrenal glands are also 100% involved in this as well

finasteride did effect 5ar type 3 though, as the info mew posted says finasteride potently inhibits it, and it is all over our brains

I would expect it to be highish, as only a portion of progesterone gets reduced to allopregnanolone.

That is a whole nother bag of crisps. Which could be very well highly significant for people with severe brain fog.

I feel like we are circling the wagons with this whole stuff, I really pray Awor’s update on Nuerosteroid study will be telling.

I would be curious to see people’s estradiol to progesterone ratio levels as well

Metformin

vancouversun.com/health/Diab … story.html

So it looks like the decrease in testosterone is dose dependent. Maybe 500mg a day to start.

ncbi.nlm.nih.gov/pubmed/22607767

I would get some metformin but i have been blacklisted by customs.

becarful Metformin just does the same thing what fin has done. I have posted about it just please do some search on the forum.
I read the case of guy who used for few days and never got back, he used clomid for months and then clomid stopped working.he was on moesso forum.

Dgreene this a great observation in regards to SSRI suffers, what they call PSSD. I believe that there is a serious serotonin connection here. My reason for thinking this

  1. Serotonin has the ability to stimulate 5ar genes expression. ncbi.nlm.nih.gov/pubmed/15936112

  2. Serotonin has the ability to increase Allopregnanolone in the brain. sciencedaily.com/releases/19 … 061714.htm

  3. My brain fog was completley abolished by a 1month treatment with 5htp(serotonin precurosor)

Although I took Paxil for years and it did not touch my brain fog. Paxil does not actually increase serotonin levels it only keeps serotonin in the synapse longer. MDMA also targets the Serotonin receptors.

you used 5HTP but did not get sexual benefits. are you planning to use 5HTP again?

I did get sexual benefits from 5htp. Higher libido, better erections and more ejaculation volume unfortunatley the sexual benefits stopped once i got off. I can’t take 5htp right now because I just got off of Paxil and am suffering from withdrawal and am extremely sick. In the future I plan to try it again. Brain fog symptoms however did not fade even after being off.

Guys check out Second Amendment right now, he is following “light at the end” treatment protocol, or a little modified version. A two prong attack of both selegeline or possibly 5htp with progesterone. IMO, this is absolutely unquestionably where all the problems start with us.

BTW, because of this storm, I drank 14 vodka & sodas on Friday night, I have had more libido and erections in the last two days than in the previous 7 months

Progesterone blog

andlos.blogspot.co.uk/2010/09/pr … id-of.html

Ok this is something, 2 people with highish progesterone levels. Getting mine checked this week. Anyone else? I have read that high progesterone can desensitize and down regulate progesterone receptors, and perhaps the highish levels come from the inhibiting effects of finasteride with regards to the progesterone to allopregnanalone conversion.

In the article written by the doctor he states to take a 100mg of progesterone day. Would that be pill form???
