Methylation Cycle and The P450 Systems

Unfortunately not at the moment but i am working on it. You could go through Wikipedia looking at the individual CYPs i mentioned. What i did was to create a list of foods that i regularly eat and then search on PubMed. The same proccess i do for supplements that i never tried and i am about to order. Lengthy process but there is no other way to be sure.

Here is my latest update :

First of all, in terms of erections quality i am back to 100% i am really happy with consistency and rigidity of my erections. Never been like that for a very long time.

-i started taking a combination of T4/T3 and i believe this is what gave ā€œthe extra edgeā€ on my erections. T3 also induces 3beta-HSD, i feel even more great, more confident, more outgoing and social. Why i decided to take T4/T3 i will explain shortly.

These are the good news. Moving on to the bad news :

-It appears that Forskolin although it worked very well, it lost its efficacy. The latest blood results have shown a return to subclinical hypothyroidism (hence me taking now thyroid medication) and my Free T to the lowest normal of the range. So Forskolin has to be cycled. 3beta-HSD Boosters are : Forskolin, All-trans Retinoic Acid with Vitamin D3, HCG. So these could be cycled and maybe there will be no problems (just an idea, i will try and report back).

Now for the most interesting part. Since my 3beta-HSD is working much better now ( i now have Perfect erections, oily skin on my forehead and oily hair and perhaps my hairline started receding a bit) my neurological side effects came back. As already discussed because of the fact that i keep a very detailed Log (for this i use an app called ā€œMy Log Proā€ for Android which i highly recommend) and a specific pattern happens every time. So now i have perfect erections but anorgasmia and also constipation has kicked in.

The more time passes by i understand what is meant by ā€œandrogen resistanceā€. More Testosterone (or better use of Testosterone from our receptors) generates oxidative stress which in turn creates several other problems.

  1. Good erections but Neurological / Psychological side effects
  2. No Neurological Side effects but Loss of Erections / Libido

To answer a couple of questions from Jonson and Airborne

Regarding the use of sulfur : Be very cautious about it. I suggest that you try and see what happens. It may or may not help you. For me drinking red wine has always been a problem (wine contains Sulfur dioxide).

Regarding my regimen : Again, i see that whatever works appears to be a very personal thing. What i can say for me is that TMG, Metafolin, Zinc, Selenium, K2, D3 seem to be working but i need more time to see this . I haven’t changed my regimen much, apart from the fact that i do not take Forskolin now.

I will wait until my TSH stabilizes and i will have new Blood work in about a month from now. I would be very interested to have the methylation panel done, its been about 6 months since i started the methylation regimen (Metafolin, TMG) and so i would like to see how things are going with my methylation. As always, i stay well away from P450 inhibitors.

I think you’re on to something here. I’ve also seen a pattern where I can have great erection, but be plagued with mental side effects. When I have more mental clarity (after getting on thyroid medication) my libido has gotten worse. It seems like I have to choose between a sex life and the ability to think clearly.

What cycle would you suggest for Forskolin? I’m about to finish the first 90-capsule bottle, one 250mg capsule a day
, 50mg active F per capsule.

Again this is trial and error. I would try 3 days on, one day off to see what happens.

Unfortunately nowadays, since 1-2 years my erections/libido aren’t amazing either.

Quick update : I am still having very good -100%- erections. I might have also found a way for stopping neurological side effects but this could be a temporary thing and so once again i will need more time to confirm this. Here are some things that although seem to be anecdotal i would like to check them out with other PFS sufferers :

1. Weather : Have you noticed getting worse before or shortly after abrupt weather changes? (Strong Winds, Sudden drop/rise of Barometric pressure of more than 2-3 Millibars). If possible use Wunderground to see how Baro/Wind changes are taking place and cross-check them with your symptoms. For me there is a very clear pattern of induction of neurological side effects before any abrupt weather change (deterioration). Have you noticed getting worse when winter comes and feeling much better during the summer?

2. Try taking Vitamin K2 + L-Carnitine together. Do you experience better erections afterwards? For me there is a very beneficial response after taking these supplements together

3. Have you noticed becoming much worse (more neurological side effects) after intensive cardio workout? For this i am not so sure but would like to check it out nevertheless

My mood is worse when weather is cloudy, I get less motivated. This is true even if I just woke up and haven’t yet looked outside to know if weather is cloudy or not.

Thought that was weird too.

I thought you had managed to stop the neurological side effects by suplementing the methylation cycle?! Is this not true anymore?

Yes the neurological sides have stopped but kicked in again when i boosted 3beta-HSD (which in turn gave me perfect erections). Now i have almost no side effects but i will have to wait to see that this lasts over time and also to have a look at Testosterone and Thyroid function. IF i continue to be like this i would say that i’m cured as i hardly now feel any side effects and Barometric changes do not affect me.

I made some changes in my regimen and more specifically :

-Switched from TMG to Choline
-Added Lion’s Mane because of its ability to protect neurons from ROS. This was also discussed in one of my earlier postings about Lion’s Mane and its ability to induce NGF.
-Added P5P
-Added CoQ10 coenzyme

User Costa suggested that i try a special form of Choline (alpha-GPC, choline alfoscerate) which i will try for sure. For the record here is my regimen :

08:00 : T3/T4 combination 50 mg
<NO FOOD, COFFEE FOR 1 HOUR because of Thyroid Med>
09:00 : Choline, Zinc/Cu, Selenium, Magnesium,Metafolin, P5P
12:00 : K2, L-Carnitine, CoQ10, D3, Lion’s Mane 3 tablets
15:00 (lunch) : Metafolin, Choline
20:00 : K2, L-Carnitine, ALA
23:00 (dinner): Metafolin, Choline, Magnesium

Dosage, per tablet :

Metafolin : 800mcg
Choline: 350mg
Magnesium: 150 mg (as Citrate)
K2 - MK7 : 100μg
CoQ10 : 30mg
L-Carnitine: 500mg

If you continue well, will you try to cut down on the supplements like…in 6 months? Or you plan to keep taking them?

Will you re-do all your exams? If you redid hormonal panel and values are corrected then we could say this is a cure! That would be great!

I just ordered p5p, and activated b12 and b9, plus I ordered vitamin D3 since I’m living in a dark country. I will be getting them this week and will be letting you know the results.
What I want is to fix my insomnia, better mood, better penile sensitivity, better libido!

Yes i will try to cut out supplements one by one to go an absolute minimum. The way that i see it now, i doubt that my thyroid function and Testosterone will ever return back to normal (i thought that Forskolin would keep things within normal range but it didn’t). OTOH if i can keep what i have now i am able to live a normal life. Yes i plan to have exams again in about a month from now.

have you tried iodine supplements for your thyroid? E.g. Nascent Iodine, Lugol’s etc.
After three months the effect of Forskolin is waning for me, I wonder how long do I need to stay out of it before the next cycle.
Also, which form of CoQ10 are you taking? When I tried Jarrow Ubiquinol QH I had a mini crash after a few days so I stopped taking it.
I guess Choline is not from Soy Lecithin.
Any recommendation for the mineral supplements (Zinc, Cu, Selenium)?

No i haven’t tried those, i will ask my endo about these supplements because i believe that iodine could also have negative effects to hypothyroid patients.

For Forskolin i would wait for 10 days then restart (since you’ve been taking it 3 months straight). The CoQ10 i am taking is from Solgar (Kaneka-Q10 - ubiquinone). The choline is also from Solgar (Choline as Bitartrate)

The Zinc/Cu formulation is from Quest (Synergistic Zinc with Copper).

I’m also weening off trazodone which I was taking for nor more than 2 weeks.

Good Find. FYI Progesterone is a DHT inhibitor. Pregnenolone could be the way to go.
