I have read people mentioning having the negative side effects of propecia worsen from stopping usage, is it possible for this to happen from one missed dose? I have had some trouble with watery semen and slightly decreased libido in recent times (but nothing big), but I recently missed one dose (yesterday) but I took it this afternoon and noticed it took me a bit longer to achieve an erection tonight, and had more difficulty ejaculating.
Highly unlikely that if you miss a dose you will have worse side effects – if anything, the opposite.
The drug is extremely potent and as such, is designed to work despite missing a dosage or two. That’s why it can take up to 7-14 days for DHT to return to baseline after a single dose – you have to wait for 5AR2 to be re-synthesized by the body to produce DHT again, and it takes awhile for this to happen.
Finasteride binds to the 5AR2 enzymes so tightly that it is an essentially irreversible process for those 5AR2 enzymes that have been bound. As such, Finasteride is extremely potent and DHT can remain significantly surpressed for days, not hours, due to amount of time it takes for 5AR2 to be re-synthesized.

You’re playing with fire here. If you are concerned enough to be on this site you should get off the drug.
